Survey: The Ideal Reference Sample for Array CGH

Introduction

Every array CGH experiment requires a normal DNA (the "Reference Sample") that is labelled up and co-hybridised to the array along with your patient sample. EuroGentest is working to make available a universal reference sample for diagnostic array CGH. To help with this, we would like to know what you would consider the ideal reference sample. Please help us by taking a few minutes to answer the questions below. The results will help us produce a reference sample which best suits the needs of the aCGH testing community.

1. Do you currently perform aCGH for diagnostic purposes?
2. Which aCGH platform(s) do you use? Check all that apply.
3. What do you currently use as a reference sample for aCGH?
4. Have you had any problems using this reference sample? e.g. Degradation, batch-to-batch variability, shortage of supply, change of source, any other problems?
If yes, please describe the problems brieflyquestion_4
5. We list below some possible characteristics of an ideal reference sample for aCGH. Please score them from 1 to 5, 1 being not important and 5 being essential
5.1. Consistent DNA Quality
5.2. Long-term availability
5.3. All CNVs documented
5.4. Derived from a European
5.5. Reasonable price
5.6. Widely used in diagnostic aCGH
5.7. CE marked for diagnostic use
6. Briefly describe other characteristics you would wish to see in your ideal aCGH reference sample
7. Do you currently use pooled DNAs as a reference sample?

If Yes, please briefly describe the pooled sample you use (e.g. Promega DNA) and say why you prefer a pooled sample over an individual sample.
If No, please say briefly why you prefer an individual sample over an pooled sample

8. Would you use a reference sample based on pooled DNAs if a suitable one became available? Please score your answer from 1 to 5 (1 being not likely and 5 being definitely)
9. If a sample with the all characteristics you consider important became commercially available at a reasonable price, how likely would you be to switch over to using it? Please score your answer from 1 to 5 (1 being not likely and 5 being definitely).
10. If you answered anything other than 5 (definitely) in Question 8, please tell us why you would wish to continue using your current reference sample (or conversely, why you would not wish to use the 'ideal' sample).

That's all - thank you!
